The National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) requires the development of a custom Single-Domain Antibody (sdAb/VHH) targeting the extracellular domain of the human Calcium-Sensing Receptor (CASR). This antibody will be used to create an injectable bioreagent for parathyroid tumor Positron Emission Tomography (PET) scanning, imaging, localization, and potentially as a radiopharmaceutical treatment for inoperable parathyroid cancer. The contractor will be required to deliver 0.5 to 1 mg each of four purified, validated, and 3XFLAG epitope-tagged sdAb (VHH) clones. Additionally, four DNA minipreps corresponding to independent sdAb (VHH) clones and complete protein and cDNA sequence information for the sdAbs must be provided. The Statement of Work outlines specific task areas including library panning, ELISA plate screening, additional services such as solution panning, negative selection, DNA synthesis, protein expression, and flow cytometry screening for validation. The contract will be a Firm Fixed Price agreement with a period of performance of DOA + 6 Months.
